Niko is now working for the head mob boss in the area (who also has connections to the shady government agent guy), and also a lower down mob boss. The head mob boss says that if Niko helps him he will bring in the last remaining guy who possibly betrayed Niko during the war, while the lesser mob boss is just giving him tons of money.
Also there is a sub plot that Niko's cousin is getting married, but that only seems to come up randomly after several missions, it may be related to how much money Niko has or maybe just scripted for a certain number of missions.
Looks like the end of the game is drawing near, there's not many eole left to kill and not much higher to go in organized crime.
